Opioid Peptide Synthesizing Enzymes

PCSKI (Prohormone convertase 1/3) is the major processing enzyme responsible for the generation of biologically active endogenous opioid peptides (and others) from their respective prohormones. This grant will generate a series of mutations in the mouse Pcsk1 gene with missense changes in protein coding amino acids of the PCSK1 gene that mimic known disease associated human SNPs in prohormone convertase 1/3. The resulting mice will be comprehensively phenotyped for metabolic, endocrine and behavioral alterations.
